目前,大環環肽化(hua)合物主(zhu)要(yao)通過以(yi)下方法進行(xing)合成:

1)各種縮合反(fan)(fan)應(ying)及取代反(fan)(fan)應(ying):如酸胺(an)縮合,sn2反(fan)(fan)應(ying)(frost,j.r.,scully,c.c.g.&yudin,a.k..nat.chem.2016,3,1105.),snar反(fan)(fan)應(ying)(spokoyny,a.m.etal.j.am.chem.soc.2013,135,5946.)。

2)金屬(shu)催化的轉化:如疊氮一炔烴環加(jia)成反應(ying),烯烴關環復分解反應(ying)(kimy.-w.,grossmannt.n.&verdine,g.l.nat.protoc.2011,6,761.),這些反應(ying)可以(yi)用于訂(ding)書(shu)肽的合成。

通過上(shang)述(shu)(shu)現(xian)有(you)的(de)(de)(de)大環(huan)(huan)化(hua)(hua)方法(fa)可以制備一系列環(huan)(huan)肽(tai)化(hua)(hua)合物(wu)(wu)(wu),以及用(yong)于穩定具有(you)特定氨基(ji)酸序列多肽(tai)的(de)(de)(de)二級結構(gou),但(dan)是現(xian)有(you)方法(fa)在成環(huan)(huan)過程中(zhong)需(xu)要(yao)依靠(kao)氫鍵等弱相互作用(yong)來實現(xian),不能直(zhi)接控制產(chan)物(wu)(wu)(wu)的(de)(de)(de)三維結構(gou)。綜上(shang)所述(shu)(shu),環(huan)(huan)肽(tai)作為(wei)一類重(zhong)要(yao)的(de)(de)(de)化(hua)(hua)合物(wu)(wu)(wu)近些年受到學(xue)術界和藥物(wu)(wu)(wu)化(hua)(hua)學(xue)領域科學(xue)家的(de)(de)(de)重(zhong)點關注,但(dan)是現(xian)有(you)合成方法(fa)的(de)(de)(de)局限性(xing)制約了新型大環(huan)(huan)環(huan)(huan)肽(tai)化(hua)(hua)合物(wu)(wu)(wu)的(de)(de)(de)合成。



技術實現要素:

本發明的(de)目(mu)的(de)之(zhi)一在(zai)于提供一種簡單實用的(de)方法,可以從(cong)簡單的(de)線性多肽前體出發,通過關環(huan)反應,制備(bei)環(huan)肽;目(mu)的(de)之(zhi)二在(zai)于制備(bei)得(de)到的(de)環(huan)肽由苯環(huan)支撐,不依靠(kao)分子內的(de)氫鍵,結構新穎。

1.一種(zhong)通過鈀催(cui)化(hua)(hua)的分子內c-h鍵的芳(fang)基化(hua)(hua)反(fan)應,高效制備新(xin)型(xing)環肽化(hua)(hua)合(he)物的制備方(fang)法,其特征在(zai)于該方(fang)法的具體步驟(圖(tu)2)。在(zai)反(fan)應瓶中依次加(jia)入原料、銀鹽(yan)、金屬鈀催(cui)化(hua)(hua)劑、添加(jia)劑(additive)以及溶劑,指(zhi)定溫度(du)下攪拌(ban)12小時,冷卻至室溫,加(jia)入乙酸乙酯稀釋(shi),硅藻土(tu)過濾,濃(nong)縮,加(jia)入二氯(lv)甲烷,飽(bao)和碳酸氫(qing)鈉水(shui)溶液洗滌,有機相用無水(shui)硫酸鈉干燥(zao),柱層析(xi)分離得(de)目(mu)標產物。

2.本發明所涉(she)及(ji)到的(de)金(jin)屬(shu)催化劑(ji)是金(jin)屬(shu)pd相關催化劑(ji),可(ke)(ke)以(yi)是pd(oac)2,也可(ke)(ke)以(yi)是其他二價pd金(jin)屬(shu)催化劑(ji)。

3.本發明所涉及到的銀(yin)鹽(yan)可(ke)(ke)以是碳酸(suan)銀(yin),醋(cu)酸(suan)銀(yin),三(san)氟乙酸(suan)銀(yin),也可(ke)(ke)以是其他一價銀(yin)金屬催化劑。

4.本發(fa)明所(suo)涉及到的(de)添加劑(additive)是(shi)羧(suo)(suo)酸相關添加劑,可以(yi)(yi)是(shi)2-苯基苯甲酸,也可以(yi)(yi)是(shi)如(ru)圖3所(suo)示的(de)其他(ta)羧(suo)(suo)酸,催化劑的(de)用量一般(ban)為0.5equiv。

5.本(ben)發明所用溶劑(ji)是叔(shu)丁醇,其用量是為(wei)每毫摩爾(er)原料(liao)對應使(shi)用范圍為(wei)40ml至200ml。

6.本發明所涉(she)及到的反應溫度可(ke)以在80℃至120℃中的任意溫度下進(jin)行。

7.本發(fa)明所(suo)用(yong)原料由氨基酸經酸胺縮(suo)合(he)得到,但不局限(xian)于(yu)這些組成片段。

本發明的優點是:

1.本發明所用各種試劑均(jun)可商業所得,原料來源廣(guang)泛,價格低廉,且各種試劑常溫(wen)常壓(ya)下能夠穩(wen)定存在,操作處理方便,無須特殊處理。

2、本發明濃(nong)度范圍為5-25mm,不需要高(gao)度稀釋,適合大量生產。

3.本(ben)發明操作簡便(bian),不需(xu)要(yao)隔絕空氣和水,一(yi)步反應即可得到兩個產(chan)物(wu)。對設(she)備要(yao)求簡單,后(hou)處理(li)也無(wu)特別要(yao)求,大(da)大(da)降低(di)了(le)合(he)成該類(lei)化(hua)合(he)物(wu)的(de)生產(chan)成本(ben)。

4.本(ben)發明(ming)所(suo)使用的催化(hua)劑用量較低,在保持(chi)良好催化(hua)效果(guo)的同時,達到了簡化(hua)工(gong)(gong)藝(yi)、降低成本(ben)、方(fang)便(bian)后處理工(gong)(gong)序(xu),溶劑的回收利用便(bian)捷,減少(shao)環境污染(ran)等要求。
